What do you do with the glowing balls? How do they help? And what causes some of the balls to disappear sometimes. I will be playing and all of the sudden a whole row will disappear or a certain color will disappear.

If they're glowing, it means that it's possible to connect 3 in a row in air. When you connect those in air, connected colour disappears, leftovers drop and "blow up" balls below.
for 5-6 long strings it's good to learn patterns. for example: right,down,right,down,right for string looking like xoxoxo. right,down,left,down,down for ooxxoo and oxxoo and ooxxo. right,right,up,right,down for ooxxo....and reversed and mirror etc. Some strings are hard to connect in air, especially if the last ball is not connecting colour.
